Detection Principle: Sandwich ELISA Method
The Human HMBS ELISA Test Kit uses a two-step sandwich enzyme-linked immunosorbent assay (ELISA).
Coating: Microplate wells are pre-coated with a Human HMBS-specific antibody.
Sample Incubation: Samples or standards are added, and HMBS binds to the immobilized antibody. After incubation and washing, only bound antigens remain.
Detection Antibody Addition: Horseradish peroxidase (HRP)-conjugated detection antibody is added, forming an antibody–HMBS–antibody “sandwich” complex.
Signal Development: TMB substrate is added. HRP catalyzes the conversion of TMB to a blue product, which turns yellow after acid stop solution is applied.
Measurement: Absorbance is measured at 450 nm using a microplate reader. The intensity of yellow color is directly proportional to the concentration of Human HMBS in the sample.
