Principle of the Assay
This kit is based on a sandwich enzyme-linked immunosorbent assay (ELISA) principle. An antibody specific for Mouse 6-keto-PGF1α has been pre-coated onto the microplate. Standards and samples are pipetted into the wells, and any target antigen present is bound by the immobilized antibody. After washing, an HRP-conjugated detection antibody is added to form an antibody-antigen-antibody “sandwich” complex. Following another wash, a TMB substrate solution is added, which develops a blue color proportional to the amount of bound 6-keto-PGF1α. The reaction is stopped with acid, turning the solution yellow. The optical density (OD) is measured at 450nm, and the concentration is calculated from the standard curve.
