Key Features & Benefits
- Optimized for Complex Matrices: The included sample diluent is specially formulated to reduce background interference from common serum/plasma components like hemoglobin and lipids, ensuring stable, low-background readings and improved data consistency.
- Streamlined One-Step Incubation: The assay features a simultaneous incubation step where the sample antigen binds concurrently with the immobilized capture antibody and the enzyme-labeled detection antibody, significantly reducing total assay time and simplifying the workflow.
- Wide Dynamic Range: Accurately quantifies S1P across a physiologically and experimentally relevant range of 50 to 2400 nmol/L.
- Excellent Specificity & Precision: Demonstrates minimal cross-reactivity and delivers reproducible results with intra- and inter-assay coefficients of variation (CV) of less than 15%.
- Exceptional Value: Achieve high-quality data affordably. Enjoy a competitive retail price of $120, or unlock greater savings with our bulk pricing, which starts under $100 per kit.
- Comprehensive Support: We offer custom ELISA kit development services and a complimentary sample testing service to meet specialized research requirements or validate the assay with your samples.
Assay Principle: One-Step Sandwich ELISA
This kit utilizes a sensitive one-step sandwich enzyme-linked immunosorbent assay (ELISA):
- Capture: Microplate wells are pre-coated with an antibody specific to Human Sphingosine-1-phosphate (S1P).
- Simultaneous Antigen-Antibody Binding: Diluted samples or standards (recommended 1:5 dilution for serum/plasma) are added to the wells along with the Horseradish Peroxidase (HRP)-conjugated detection antibody in a single step. S1P present in the sample is captured between the two antibodies.
- Signal Development: After washing to remove unbound material, a Tetramethylbenzidine (TMB) substrate solution is added. The HRP enzyme catalyzes the conversion of TMB to a blue-colored product.
- Quantification: The reaction is stopped with an acid solution, changing the color to yellow. The absorbance (Optical Density, OD) at 450 nm is measured. The intensity of the yellow color is directly proportional to the concentration of S1P in the original sample.

Workflow & Sample Preparation Note
Key Pre-Assay Step: For serum and plasma samples, a 1:5 dilution is required. Combine 10 µL of sample with 40 µL of the provided Specialized Sample Diluent. This step is crucial as the diluent mitigates the effect of interfering substances, ensuring assay accuracy and consistency.
*Final concentration = Calculated concentration from standard curve × 5 (dilution factor).*
Basic Protocol Overview:
- Add 50µL of standards or diluted samples to wells.
- Add 100µL of HRP-conjugated Detection Antibody to each well (except blank).
- Incubate at 37°C for 60 minutes.
- Wash plate 5 times.
- Add TMB Substrate (A&B), incubate at 37°C for 15 min in the dark.
- Add Stop Solution and read absorbance at 450 nm.

Frequently Asked Questions (FAQ)
Q: Why is a 5-fold dilution required for serum/plasma?
A: The dilution step with our specialized diluent is critical. It reduces the concentration of matrix interferents (e.g., lipids, hemoglobin) that can cause high background or unstable signals, thereby improving assay specificity, accuracy, and reproducibility.
Q: What are the benefits of the one-step incubation?
A: The single, simultaneous incubation of sample and detection antibody reduces total hands-on time, minimizes pipetting steps (and potential errors), and shortens the overall time to results, enhancing lab efficiency.
Q: How do I qualify for bulk pricing under $100?
A: Simply contact our sales team directly. Pricing below $100 per kit is available for orders of multiple kits. We provide tailored quotes based on your specific volume needs.
Q: Is this kit suitable for clinical diagnostics?
A: No. This product is developed and validated for Research Use Only (RUO). It is not intended for diagnostic procedures.
