Principle: Both kits use a One-Step Sandwich ELISA. The target NELL1 in samples is captured between a pre-coated antibody and an HRP-conjugated detection antibody. After washing, TMB substrate produces a color change measurable at 450nm, with intensity proportional to NELL1 concentration.

Sample Handling & Workflow
Collection: Follow standard protocols for serum, plasma (EDTA/citrate/heparin), cell culture supernatant, or tissue homogenate. Centrifuge at 3000×g for clarification.
Storage: Aliquot and store at -20°C; avoid repeated freeze-thaw cycles.
General Procedure:
- Add 50µL of standards or diluted samples (10µL sample + 40µL diluent for Tier 1 serum/plasma) to wells.
- Add 100µL of the respective HRP Detection Antibody.
- Incubate at 37°C for 60 min.
- Wash plate 5x.
- Add TMB Substrate (A&B), incubate at 37°C for 15 min (dark).
