Assay Principle: Sandwich ELISA

This kit employs a solid‑phase sandwich enzyme‑linked immunosorbent assay. The microplate is pre‑coated with a monoclonal antibody specific for Mouse IL‑6. Standards and samples are added, allowing IL‑6 to bind to the immobilised antibody. After washing, a Horseradish Peroxidase (HRP)‑conjugated detection antibody is added, forming an antibody‑antigen‑antibody “sandwich”. Following a second wash, TMB substrate is added. The HRP enzyme catalyses a blue colour that turns yellow upon acidification. The optical density (OD) at 450 nm is directly proportional to the IL‑6 concentration in the sample.

Sample Dilution Note: For serum or plasma, samples are pre‑diluted 5‑fold during the procedure (10 μL sample + 40 μL diluent). Multiply final results by 5.

StorageExtract as soon as possible. Store at –20 °C (≤ 1 month) or –80 °C (≤ 2 months). Avoid repeated freeze‑thaw cycles.

Important: Do not use samples containing sodium azide (NaN₃), as it inhibits HRP activity.


Materials Required But Not Provided

  2. Precision pipettes and tips (0.5–10 μL, 2–20 μL, 20–200 μL, 200–1000 μL)
  3. 37 °C incubator or water bath
  4. Distilled or deionised water

Procedure Notes & Important Tips

  1. Bring all reagents to room temperature before use. Concentrated wash buffer may crystallise; dissolve crystals in a warm water bath.
  2. Unused strips must be resealed with desiccant and stored at 2–8 °C.
  3. Standard dilution: Prepare serial dilutions of the stock standard according to the lot‑specific datasheet.
  4. Sample addition: Add 40 μL sample diluent to each sample well, then 10 μL test sample. Mix gently without touching the well bottom.
  5. Incubation: Use the plate sealer and incubate at 37 °C for 60 minutes (first incubation).
  6. Washing: After each incubation, discard liquid, fill wells with wash buffer, stand for 30 seconds, then discard. Repeat 5 times. Incomplete washing may cause false positives.
  7. Substrate: Add Substrate A then B. Incubate in the dark at 37 °C for 10–15 minutes.
  8. Read OD: Add stop solution and read at 450 nm within 15 minutes.

Detailed Assay Procedure

  1. Remove required strips from the foil pouch after 20 min equilibration. Reseal unused strips.
  2. Standards: Add 50 μL of each standard concentration to standard wells.
  3. Samples: Add 40 μL sample diluent to sample wells, then 10 μL test sample. Blank wells: leave empty.
  4. Add 100 μL HRP‑conjugated detection antibody to all wells except blanks. Seal and incubate at 37 °C for 60 min.
  5. Aspirate, wash 5 times (manual or automatic washer).
  6. Add 50 μL Substrate A and 50 μL Substrate B to each well. Gently tap to mix. Incubate at 37 °C in the dark for 10–15 min.
  7. Add 50 μL stop solution. Read OD at 450 nm within 15 min.

Data Analysis

Plot standard curve: OD values (y‑axis) vs. standard concentrations (x‑axis) using linear regression (R ≥ 0.9900). Calculate sample concentrations from the curve. For serum/plasma samples, multiply by the dilution factor (5×). For cell culture supernatants or other matrices, use the appropriate dilution factor as validated.
