Test Principle

This Rat Interleukin 22 (IL-22) ELISA Kit employs a double-antibody sandwich technique. A monoclonal antibody specific for rat IL-22 is pre-coated onto the high-binding microplate. Standards and samples are added, allowing IL-22 to bind to the immobilized antibody. After washing, an HRP-conjugated detection antibody that recognizes rat IL-22 is added, forming an antibody-antigen-enzyme complex. Unbound conjugate is removed by thorough washing. TMB substrate is then added; the HRP enzyme converts TMB into a blue product, which turns yellow after the stop solution is introduced. Absorbance is read at 450 nm. The color intensity is directly proportional to the concentration of rat IL-22 in the sample.

Sample Collection and Preparation

Proper sample handling preserves analyte integrity. Follow the guidance below; for full details, refer to the kit manual.

Serum – Allow blood to clot naturally at room temperature for 10–20 minutes. Centrifuge at 2000–3000 rpm for 20 minutes. Collect the supernatant promptly. If precipitate appears after storage, centrifuge again before use.

Plasma – Use EDTA or sodium citrate as the anticoagulant as recommended for your study. Mix gently for 10–20 minutes after collection, then centrifuge at 2000–3000 rpm for 20 minutes. Transfer the supernatant and re-centrifuge if any precipitate forms.

Urine – Collect aseptically in a sterile tube. Centrifuge at 2000–3000 rpm for 20 minutes. Use the supernatant. The same approach applies to cerebrospinal fluid and pleural/ascitic fluid.

Cell culture supernate – For secreted proteins, collect the culture medium and centrifuge at 2000–3000 rpm for 20 minutes. For intracellular IL-22, resuspend cells in PBS (pH 7.2–7.4) at approximately 1×10⁶ cells/mL, lyse by repeated freeze-thaw cycles, then centrifuge and harvest the supernatant.

Tissue homogenates – Weigh the tissue, add a measured volume of PBS (pH 7.4), and homogenize thoroughly on ice. Centrifuge at 2000–3000 rpm for 20 minutes, then collect the supernatant. Aliquots can be frozen at -20°C or below for later use.

Critical notes:

  • Extract and assay samples as soon as possible. If a delay is unavoidable, store at -20°C and avoid repeated freeze-thaw cycles.
  • Do not use samples containing NaN₃. Sodium azide inhibits HRP activity and will cause inaccurate readings.

Reagent Preparation

  • Bring all reagents to room temperature (15–30 minutes) before use.
  • Wash buffer: If crystals appear in the concentrated wash buffer, warm the solution in a water bath until the crystals dissolve. Dilute the concentrate with distilled water (30-fold for the 96T format, 20-fold for the 48T format) to prepare 1× working wash buffer.
  • Standards: Reconstitute as instructed in the manual and prepare serial dilutions to obtain the standard curve points: 24 ng/L, 12 ng/L, 6 ng/L, 3 ng/L, and 1.5 ng/L.

Assay Procedure Summary

  1. Add standards and samples: Pipette 50 µL of each standard into designated wells. For sample wells, first add 40 µL of sample diluent, then 10 µL of sample (5-fold final dilution). Leave blank wells empty of sample and enzyme conjugate.
  2. Incubate: Cover the plate with a fresh plate sealer and incubate at 37°C for 30 minutes.
  3. Wash: Remove the sealer, discard the liquid, and wash each well 5 times with 1× wash buffer. Soak for 30 seconds per wash, then blot dry.
  4. Add enzyme conjugate: Add 50 µL of HRP-conjugated detection reagent to each well, except the blanks.
  5. Incubate: Cover and incubate again at 37°C for 30 minutes.
  6. Wash: Repeat the 5-step wash process.
  7. Develop color: Add 50 µL of Chromogen Solution A and 50 µL of Chromogen Solution B to each well. Mix gently and incubate in the dark at 37°C for 10 minutes.
  8. Stop reaction: Add 50 µL of stop solution to each well. The color changes from blue to yellow.
  9. Measure absorbance: Read the optical density (OD) at 450 nm within 15 minutes, using the blank well to zero the reader.

Calculation of Results

Construct a standard curve by plotting the OD values against the known concentrations of the standards (24, 12, 6, 3, 1.5 ng/L). Use the resulting linear regression equation or curve to determine the IL-22 concentration of each sample. Multiply the obtained value by the final dilution factor (e.g., ×5) to calculate the actual concentration. If the sample was further diluted beforehand, also multiply by that extra factor.


Storage and Stability

  • Storage: 2–8°C for all kit components.
  • Shelf life: 6 months from the date of manufacture when stored unopened as directed.
  • After opening the pre-coated plate, return unused strips to the sealed pouch with desiccant and store at 2–8°C.
