  • Cell Culture Supernatant: Centrifuge at 3000 rpm for 10 min to remove particulates.
  • Tissue Homogenate: Homogenize equine tissue (e.g., parathyroid, bone) in normal saline, spin at 3000 rpm for 10 min, and retain the supernatant.
  • Storage: If not assayed immediately, aliquot and store at -20°C. Avoid repeated freeze‑thaw cycles. Thaw completely at room temperature and mix gently before use.

  4. Washing: Aspirate, then wash 5 times with 1× wash buffer (dilute 20× concentrate 1:20; dissolve any crystals by warming). Soak 1 min per wash, then blot dry.
  5. Substrate: Add 50 µL Substrate A + 50 µL Substrate B. Incubate in the dark at 37°C for 15 min.
  6. Stop & Read: Add 50 µL Stop Solution. Measure OD at 450 nm within 15 min. Plot the standard curve and calculate sample concentrations. Multiply the result by 5 to correct for the assay dilution.
